Collagen supplements: what digestion does to them

Two equally confident claims circulate about collagen. One promises firmer skin after a month of scoops. The other answers that protein is dismantled in the stomach, so paying extra for this particular one is pointless. Both describe digestion inaccurately, and the easiest way through is step by step.

What collagen is, and why there is so much of it in you

Collagen is the most abundant protein in the human body — roughly a third of all the protein you are made of. Tendons, ligaments, cartilage, blood vessel walls and the layer of skin that holds its shape, the dermis, are all built from it. Bone is about half collagen by volume: minerals crystallize along its fibers the way concrete sets around rebar.

Its composition is what sets it apart from other proteins. About every third amino acid in a collagen chain is glycine, the smallest one there is. Proline and hydroxyproline sit alongside it in unusual quantity, and tryptophan is absent entirely. That lopsided makeup explains both what shows up in your blood after a dose and why collagen makes such a poor protein powder.

What digestion does to it

Exactly what it does to any other protein. Collagen has no special route through the gut.

In the stomach, hydrochloric acid unwinds the triple helix and pepsin starts cutting the chains into pieces. In the small intestine, pancreatic enzymes take over, joined by enzymes anchored on the surface of the intestinal cells themselves. By the end of that sequence what remains of the protein is free amino acids and short fragments two or three amino acids long.

Those are then absorbed by the cells of the intestinal wall and passed into the blood. No "collagen from the tub" arrives in your skin: a molecule that size does not cross the intestinal wall, and the idea that swallowed collagen is bolted onto the collagen in your face contradicts the physiology.

This is where the popular debunking usually stops. The picture past that point is more interesting.

Why "it just breaks down into amino acids" is too simple

Not everything is absorbed as separate amino acids. Some short peptides cross the intestinal wall intact, and collagen produces a characteristic set of them: pairs built from proline and hydroxyproline. After a dose of hydrolyzed collagen they turn up in the bloodstream — this has been measured and reproduced in independent labs.

Hydroxyproline is almost absent from ordinary food. The body makes it itself, after the fact, by modifying proline that is already stitched into a collagen chain. That makes these peptides a usable marker: their appearance in blood shows that what was eaten really was collagen rather than protein in general.

From there researchers test a hypothesis. Those fragments may act not as building material but as a signal — passing within reach of the cells that manufacture collagen and nudging them to work harder. The fragment is not inserted into your skin. At best it reports that raw material is plentiful.

The word "hypothesis" is not a hedge here. Proving that a peptide reaches the blood is one claim, and it has been demonstrated. Proving that a person's skin or joint changes because of it is a different claim, and it holds up considerably less well.

What the human trials show

There are a lot of trials by now, and their results are uniformly modest.

Skin. Studies of hydrolyzed collagen usually measure hydration and elasticity with instruments. They do find shifts; the shifts are typically small and not present in every participant. The studies also share a set of features: they are short, most often 8 to 12 weeks, they enroll a few dozen people, and the overwhelming majority are paid for by supplement manufacturers. That does not make the results false on its own, but it explains why the conclusions are worded so cautiously.

Joints. Here the outcomes measured are pain and mobility — things the participant reports personally, which makes them the measurements most easily bent by expectation. Some trials in osteoarthritis and in athletes find a reduction in pain; others find nothing. Nobody has convincingly shown cartilage rebuilding on imaging.

Hair and nails. Markedly less data, and what exists is weaker in quality.

The honest summary is boring: collagen is neither a fraud nor a treatment. If there is an effect, it is small, it takes a couple of months rather than days, and in the results it appears as a shift in group averages rather than a transformation.

As a protein source, collagen is poor

This follows directly from the composition described at the start. Collagen contains no tryptophan and only small amounts of several other essential amino acids — the ones the body cannot synthesize and has to obtain from food.

The standard scores for protein quality put it near the bottom for that reason. Twenty grams of collagen and twenty grams of whey give the body quite different things, even though the number printed on the tub is the same. Collagen does count toward your daily protein, but as a small contribution, and there is no sense in letting it displace ordinary protein sources.

The flip side: collagen is not a harmless zero. It is protein, and it counts in the arithmetic. A 10-gram scoop is roughly 35 to 40 calories, and in ready-made collagen drinks the added sugar or juice usually contributes more calories than the collagen does.

The difference between gelatin and hydrolysate is convenience and fragment size, not "absorbed or not absorbed." Both are absorbed.

Doses in the skin and joint studies generally fall between 2.5 and 10 grams of hydrolysate per day; with native type II the numbers are in tens of milligrams. Marine or bovine makes no difference to the outcome inside you: the same fragments reach the blood either way.

And a word about the rules. In the European Union collagen is sold as a food supplement, not a medicine, which means it is regulated for safety and honest labeling rather than assessed for efficacy the way a drug is. There is no authorized EFSA health claim linking collagen intake to skin condition, so under the claims regulation a package cannot legally say that it firms your skin — which is why labels talk around it with pictures and adjectives instead. The same gap between a mechanism on the box and evidence in people runs through activated charcoal sold as a detox.

What follows from this

Collagen reaches the blood in fragments rather than whole, and its mechanism is not the one the advertising describes. The fragments may act as a signal, but the link between that signal and firmer skin in a particular person remains unproven. The effects in studies are real, they are small, and nearly all of the work was paid for by the people selling the product.

No harm has been described at ordinary doses, so if you enjoy taking collagen, take it. But between a tub of collagen and enough ordinary protein on the plate, the plate delivers more and costs less. And a supplement bought instead of a medical appointment for joint pain costs far more than its price: it consumes the months during which the joint is going untreated. Persistent joint pain, swelling, or a joint that locks or gives way is a reason to see a doctor, not to increase the scoop.

Can I replace the supplement with gelatin, bone broth, or jelly?
By composition gelatin is the same collagen, so in that sense yes. The difference is in how precisely you can dose it and in what comes along with it: fat in a rich broth, sugar in a ready-made jelly. Bone broth is also variable — the collagen content depends entirely on how it was made.
Should I take collagen with vitamin C?
Vitamin C really is required to assemble your own collagen, which is why it is often included in the blend. But it works as insurance against deficiency, not as an amplifier: an ordinary diet supplies enough, and extra beyond that does not speed anything up.
